A Prickly Subject: Apoptotic Regulation by Hedgehog Morphogens
نویسنده
چکیده
Morphogens, as intercellular signalling proteins, provide a non-cell-autonomous mechanism to impart positional information to cells and govern essential cellular processes such as apoptosis. Individual morphogen pathways utilise diverse strategies to regulate the assembly and activity of the pro-apoptotic multi-protein complexes – namely the apoptosome and the death-inducing signalling complex (DISC). This review aims to highlight the apoptotic regulatory mechanisms utilised by the Hedgehog (HH) morphogen pathway – with particular emphasis on a novel Caspase-9 activating complex and the utilisation of a pro-apoptotic autocrine-signalling loop.

Hedgehog and Decapentaplegic instruct polarized growth of cell extensions in the Drosophila trachea.
The migration of cellular extensions is guided by signals from tissues with which they contact. Many axon guidance molecules regulate growth cone migration by directly regulating actin cytoskeletal dynamics. Secreted morphogens control global patterns of cell fate decisions during organogenesis through transcriptional regulation, and constitute another class of guidance molecules. We have inves...
متن کاملA fixation method to preserve cultured cell cytonemes facilitates mechanistic interrogation of morphogen transport
During development, extracellular cues guiding cell fate determination are provided by morphogens. One mechanism by which morphogens are proposed to traverse extracellular space is by traveling along specialized filopodia called cytonemes. These cellular highways extend between signal-producing and -receiving cells to enable direct morphogen delivery. Although genetic studies support cytoneme i...
متن کاملReggie-1/flotillin-2 promotes secretion of the long-range signalling forms of Wingless and Hedgehog in Drosophila
The lipid-modified morphogens Wnt and Hedgehog diffuse poorly in isolation yet can spread over long distances in vivo, predicting existence of two distinct forms of these morphogens. The first is poorly mobile and activates short-range target genes. The second is specifically packed for efficient spreading to induce long-range targets. Subcellular mechanisms involved in the discriminative secre...
متن کاملTrafficking, development and hedgehog
Embryogenesis is mediated by a relatively small number of developmental signaling pathways, and the morphogens, receptors and transcription factors integral to these cascades are considered the master regulators of development. However, superimposed on this is an additional layer of control by complex intracellular trafficking networks. The importance of trafficking in controlling the processes...

The mechanisms by which morphogens, such as Sonic hedgehog (Shh), specify distinct cell fates in a concentration-dependent manner are not fully understood. Shh signaling is regulated by a feedback network that comprises Shh-binding factors, the expression of which is controlled by the Hedgehog pathway itself. Recent studies have identified the hedgehog-binding protein growth arrest-specific gen...
